I'm looking at draft to weave a set of simple, plain weave hand towels. The project suggests a 12 EPI with a Reed 8, sleyed at 1,2 /dent or a Reed 6 sleyed at 2/dent. Would it matter if I used a Reed 12 sleyed at 1/dent? Is there an advantage/disadvantage to any of these or is the idea to get to a desired EPI with the reeds you have available?

I depends on what you're using for warp.  If the warp is thick enough that it doesn't fit well in the dents of the 12 reed, it would make a difference.  If you are using something like 5/2 cotton, it wouldn't.

With an irregular sleying pattern such as the 8 dent reed or the doubled threads in a 6 dent, you will get some unevenness in the cloth corresponding to the uneven spacing of the warps in those reeds.   These often wash out, especially in cotton toweling.  But as long as you have a 12 dent reed and the yarn slides easily in the dent it is in, you can use that and get the most even spacing.

  Many times you have to compromise with a reed that you have and the epi that you want to accomplish.  Any irregularities are not distracting when they are consistent, anyway.

12 should be fine. I think the alternatives were for if you didn't have a 12.
